When choosing, check the maximum relative centrifugal force (x g) the tube is rated for against your protocol and rotor; ratings differ between makers and between conical and self-standing designs. Polypropylene suits most aqueous work and resists many solvents, while polycarbonate bottles are clearer but less resistant to some chemicals. Decide between sterile tubes packed in racks or bags and lower-cost bulk non-sterile packs, and between attached and separate caps. Replacement caps for some 15 mL and 50 mL tubes are sold on their own, and a 50 mL centrifuge tube filter is available for sterile filtration.
A centrifugation, or centrifuge, tube is a tube made to withstand the force of spinning in a centrifuge so that solids, cells or denser liquids separate from the rest of the sample. Most disposable tubes are polypropylene with a screw or snap cap, ranging from microtubes of 2 mL or less to conical tubes of 15 mL and 50 mL and bottles of 250 mL or more.
A conical tube has a tapered bottom that collects a pellet in a small point, which makes the pellet easier to see and the liquid above it easier to remove. A self-standing, or skirted, tube has a base that lets it stand upright on the bench without a rack. Self-standing tubes are convenient for storage and handling, but check their g-force rating and fit in your rotor.
It depends on the tube's size, material and design, so use the maximum RCF stated by the manufacturer rather than a general figure. Exceeding the rating can crack or deform the tube and cause leaks inside the centrifuge. Also fill tubes within the recommended volume, balance the rotor, and use adapters made for that tube size.
